APMA activation of Matrix Metalloproteinases  

1.    Make a fresh 10mM stock of APMA ( 4-amino­phenyl­mercuric acetate) using 0.1M NaOH  (This must be done prior to activation protocol) 

2.    Dilute stock APMA 4x in TCNB buffer – 50mM Tris, 10mM CaCl2, 150mM NaCl, 0.05% Brij35, pH7.5 

3.    Adjust pH to 7.5 using HCl 

4.    Dilute sample 1 in 5 with APMA solution 

5.    Incubate at 37C dependant on MMP.   

6.    MMP-2 requires 2 hours whilst MMP-9 requires 5 hours incubation.   Please e-mail us times for other MMPs.
